Nobel for microscopy that reveals inner world of cells
Ever since the seventeenth century, when the early microbiologist Antonie van Leeuwenhoek focused light through lenses and marvelled at the cells that swam before his eyes, microscopes have opened up new vistas of discovery. This year, the Nobel Prize in Chemistry went to three scientists who defied the limits of light microscopes to reveal sharp images of molecular scale structures in living cells.
The advances made by Stefan Hell, William Moerner and Eric Betzig in the 1990s and 2000s mean that biologists can now see, in real time, how proteins are distributed and move inside cells at the junctions between neurons, for example, or in fertilized eggs dividing into embryos.
"It is really a revolution for the life sciences, because we can see structures that we could never see before," says Stefan Jakobs, who works with super resolution techniques at the Max Planck Institute for Biophysical Chemistry in Gttingen. Or as the Nobel committee put it: "Microscopy has become nanoscopy."
No matter how clean their lenses, optical microscopes inevitably provide a blurry view of the molecules inside cells, as German physicist Ernst Abbe realized in 1873. The laws of physics dictate that visible light cannot distinguish between objects closer to each other than around 200 nanometres (around half the wavelength replica van cleef jewelry of visible light) they will appear as one blob. Moerner went on to show that identical molecules could be made to fluoresce at different times, a discovery that ultimately made Betzig's vision a reality.
It would take a decade before Betzig demonstrated his idea in practice. He left academic science to work in his father's machine tool business in Michigan. In 2006, working at the Howard Hughes Medical Institute's Janelia Farm research campus in Ashburn, Virginia, he used the technique to take a super resolution picture of a lysosome protein dotted with green fluorescent molecules as labels. The technique can now get down to a resolution of 20 nanometres, says Markus Sauer, who studies super resolution microscopy at the University of Wrzburg, Germany.
Meanwhile Stefan Hell, working at the University of Turku in Finland, had found a way around Abbe's limit by a different technique, which also relies on switching fluorescent molecules on and off. In 1994 he proposed using one laser to make a cluster of dye molecules fluoresce, and a second beam, of a different wavelength, to switch some of those fluorophores off through a process described by Einstein in 1917.
Hell's trick is to use the second beam to outline the cluster illuminated by the first, so that only the molecules in a very narrow spot fluoresce. The final image remains blurred, as light still cannot beat Abbe's limit, but it is clear that light can have come only from the narrow central spot defined by the second beam, enabling researchers to pinpoint the light source.
Building up a series of these tiny fluorescent spots creates a fine resolution picture. "There will be many new versions of super resolution microscopes in future," Hell says.

Neighbors in disbelief over decade old Santa Ana kidnapping
Neighbors at an apartment complex in Bell Gardens, where a man is alleged to have lived with a woman he kidnapped a decade ago, were in disbelief Wednesday when they heard he had been arrested.
The neighbors at the small, two story building insisted that Isidro Garcia was a good husband who cared for his family.
The 42 year old Garcia was arrested earlier this week on suspicion of raping and kidnapping his girlfriend's 15 year old girl 10 years ago. Police allege that Garcia drugged and kidnapped the girl and beat her when she tried to flee
As the sky darkened, neighbors huddled outside their doors, all of them saying the accusations couldn't be true.
Garcia and the woman, who has not been identified, had been living in Bell Gardens for at least six years.
The woman took her 2 year old child to the Bell Gardens Police Department on Monday and accused Garcia of domestic violence.

"He was a hard working man, he worked two jobs," Ledesma said. "He would do anything for her and their daughter."

"He was a hard working man, he worked two jobs," Ledesma said. "He would do anything for her and their daughter."

I first heard about them in 2009 after I met Richard X, who produced part of their album. To be honest it difficult for me to pick out one particular song of theirs, because I really love the whole thing, but in Dresden is definitely one of my favorites. The production is great simple, energetic, and I love Eleanore Everdell vocals. I hope that I get to see them live next year.

I spent many nights listening to this song over and over trying to pick apart how what seems to be a simple song has layer upon layer of precisely placed melody and instrumentation. I think this is going to be one of the bands people will look at 20 years from now as defining part of the sound of this generation, and, more so, the feel of this generation. I loved the self confidence in the randomness of the words in both soul sister / Ain that Mister Mister on the radio / The way you move ain fair you know puts cut up lyrics to shame, and the final chant of Romance is pop genius if anything is. Despite the boasts, I get the feeling Nicki thinks the whole world is against her. She is constantly on the defense. I guess this comes with every great from pop singers to NFL players the questioning, the constant doubt, the failure forecasts from everyone watching. It a delirious swirl of arpeggiated synths and chopped, repetitious (really repetitious) vocals. On paper, seems like a pretty standard post Robyn piece of robotic synth pop, but since it cuts out all the chaff, it pretty much an immediate, seemingly unending fun spiral. I obsessed.

I can listen to music when I write. This means that during the final two years of writing my book, I hardly listened to anything at all. Then this summer Stereogum posted Frankie Rose and the Outs playing Brown Haired Girls, and I felt like my brain was a tomb being aired out for the first time in 2,000 years. There nothing extraneous in it simply explodes over and over in fiestas of balls out exuberance, then pulls back so each new verse feels like a revelation. Frankie and Co. nail this incredible dream pop/garage sound, a gauzy shawl of distortion and reverb around a core of driving determination, crowned with a gooey, oozing layer cake of vocals encrusted in spun sugar fuzz. It barreled a path clear through my
head and let the sun shine in. If I had to just pick the song that spoke to me the most personally, it My Time by Kurt Vile and the Violators. The lyrics touch me on a personal level and are very relatable. The bright production is a fresh departure for Kurt and the band; it confident and tasteful in a less is more kind of way; that guitar solo in it is right up my alley, and frankly I surprised that this song didn make a bigger splash. All the same, it was and is my favorite song of 2010.

The song that blew my mind in 2010 was Anika cover of Yoko Ono Yang. Picking up the record randomly at Rough Trade, I wasn sure what to expect, but after my first listen to the first track, I was ready to buy. Then as I listened further I fell into the second track (as all records should make you do). Yang is dirty 2010 post punk. It is distortion, reverb, electronic, dub and rhythm that could break down at any minute. Fragile.
